Artem Siemens, Miltenyi Biotec, talk at the 3rd focal Molography User Meeting 2026: Development of an Fc-Capturing Protein for High-Throughput Screening at Miltenyi Biotec

Artem Siemens (Miltenyi Biotec) shows how a homodimeric capture nanobody turns focal molography into a multiplexed screening workflow for Fc-based binders, running a 16-plex assay for 96 samples within half a day.

About the Talk

Recorded at the 3rd focal Molography User Meeting on September 9, 2026, in Adliswil, Switzerland. In this 14-minute flash talk, Artem Siemens gives a view from inside Miltenyi Biotec: how nanobodies are developed together with lino Biotech and the University of Applied Sciences and Arts Northwestern Switzerland (FHNW), how the demand for binder characterization keeps growing, and how his team built a capture system to keep pace with it.

What the recording covers

  • A capture nanobody instead of Protein A/G. A homodimeric variable domain of a heavy-chain-only antibody (VHH), immobilized by DNA-directed immobilization (DDI), captures Fc-based binders with no coupling or purification step. No dissociation was observed over a six-hour run, while Protein A/G lost signal over time.
  • Multiplexed kinetics of Fc-based binders. Immunoglobulin G (IgG), single-chain variable fragment Fc fusion (scFv-Fc) and VHH-Fc constructs measured side by side on one chip, with a free reference mologram group confirming the absence of non-specific binding. The binders differ mainly in their dissociation rate, which drives the resulting affinity.
  • Shorter runs through pre-incubation. Mixing capture nanobody and binder before injection cut the measurement time from four to three hours, and parallel injection brought it down to two hours, with affinity values in a similar range across all three setups.
  • A third pillar for throughput. Next to routine measurements and method development, the new workflow runs a 16-plex assay for 96 samples within half a day, with further capture systems in development.

About the Speaker

Artem Siemens is Team Coordinator in the Binder Discovery Group at Miltenyi Biotec. He leads the group's cloning workflows and oversees routine affinity and kinetic analysis on MACS Matchmaker, including the development of new applications on the platform.
