At the 11th Novalix Biophysics in Drug Discovery Conference (June 2025), Dr. Andreas Frutiger, Head of R&D at lino Biotech AG, presented how focal molography addresses the DEL hit validation bottleneck.
The DEL validation problem
The traditional DEL workflow requires off-DNA resynthesis and characterisation of every promising hit. This generates significant work in the chemistry department while producing limited biologically relevant information, a bottleneck that scales poorly when a selection returns tens or hundreds of hits worth pursuing.
Focal molography's contribution
By exploiting the DNA tag for directed immobilisation, focal molography enables direct on-DNA characterisation: fast and label-free kinetic profiling of up to 54 DEL hits in a single instrument run, without the need for off-DNA resynthesis. Kinetic parameters kon and koff are read directly from the on-DNA compound bound to the sensor surface.
